Locking hasn't been a feature for too long, if I remember correctly, you need at least Subversion 1.2 as base so please check if both client and server support it.

Concerning TortoiseSVN, it is an independent project and generally you should questions regarding it on the TSVN mailinglist. However, TortoiseSVN has an entry in its context menu (right click) which allows locking.
