I had the same problems on my Vista machine and i looked in the EventLog and
found the following entry:

Fehler beim Generieren des Aktivierungskontextes für "C:\Program
Files\TortoiseSVN\bin\TortoiseSVN.dll". Die abhängige Assemblierung
"Microsoft.VC90.CRT,processorArchitecture="amd64",publicKeyToken="1fc8b3b9a1e18e3b",type="win32",version="9.0.21022.8""
konnte nicht gefunden werden. Verwenden Sie für eine detaillierte Diagnose
das Programm "sxstrace.exe".

It seems that the move to VS 2007 triggered the error. But an uninstall,
reboot, install cycle installs TSBN just fine again until the next nightly.
So on a fresh install anything seems fine.
